Qantas later confirmed the plane had landed safely, but still did not explain the diversion.
However, the Australian Transport and Safety Bureau (ATSB) said the plane had some pressurisation problems mid-flight.
"It was reported that some time en route to Townsville the aircraft experienced a cabin depressurisation," a spokesman told AAP.
Flight tracking data shows the plane dropped from about 40,000ft (12,200m) to 10,000ft (3000m) in about five minutes.
The spokesman said ATSB would investigate and engineers would be on hand to assess the aircraft upon landing.
"Obviously, we'll be looking at it very closely and in due course," he said.
